Desmond Trufant was born to Constance Trufant and Lloyd Trufant on September 10, 1990.
He was born in the Washington city of Tacoma. Des Trufant is the youngest of three Trufant siblings.

In his senior year, the Seattle Times named him first-team All-State. He was also named to the Seattle Times All-State postseason team.

Season 2013 of the Atlanta Falcons
Desmond was selected by the Atlanta Falcons in the first round (22nd overall) of the 2013 NFL Draft. He was the third cornerback chosen, following Dee Milliner (overall: 9th) and D.J. Hayden (overall: 12th).
On July 24, 2013, he agreed to a four-year contract worth $8.16 million. The contract also included a guaranteed $6.94 million and a $4.31 million signing bonus.
Because the Falcons’ training camp was overrun with cornerbacks, Trufant had to compete for a spot on the team. He competed for the two cornerback positions against Robert McClain, Asante Samuel, and Robert Alford.
Falcons head coach Mike Smith was impressed with his performance and chose him to start alongside Asante.
Following that, he made his NFL debut in a loss to the New Orleans Saints in the season opener. Trufant began with one impressive performance after another.
